How fantastic to be part of a session with Betsy Crouch (UCSF), Thomas Wälchli (UCL), Alexander Ligocki (Vanderbilt), and Fiona Qiu (INSERM) with discussion leader Vanessa Coelho-Santos (University of Coimbra) at the Barriers of the CNS GRC. The future is bright! 🧠 🏰 💡 www.grc.org/barriers-of-...

📃Preprint alert @biorxivpreprint.bsky.social. Does your 2026 involve ICV injection of AAVs 🧠? If so, this study led by Luke is for you. "Adeno-associated viruses (AAVs) induce dose-dependent neonatal ventriculomegaly following intracerebroventricular administration" www.biorxiv.org/content/10.6...
Representative 3D reconstructions of brain ventricles from control and high-dose AAV2/5-injected pups at 2, 5, and 7 days post-injection (P2.5, P5.5, P7.5). Quantification of total ventricular volume across time points. (n = 3 per condition).

Dose is key🗝️. Higher doses of AAV (1uL of 4 x 10^12 GC/mL) consistently induced ventriculomegaly and permissive tropism in neonates, while moderate doses (1uL of 1 x 10^12 GC/mL) did not. Study led by Luke Liu, with true collaborative effort with Ania Noguera, Ryan Humphries, and Blake Zhou.
Representative coronal sections at the level of the posterior anterior commissure (pAC; dashed outline) showing lateral ventricle morphology in P7.5 brains following P0.5 ICV injections.
