Overall, we propose a model where the barrier formation in roots is integrated in an N-sensing mechanism that uses SGN3. This mechanism is largely hidden in standard agar-based conditions, but becomes clear in low N agriculture conditions - obviously raising an insane amount of cool questions.
120
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
Lastly, since SGN3 binds to SERK co-receptors we found that stacking mutants in theses can also uncouple CS formation and N-signaling! Further supporting a model where the SGN3 signaling depends on ligand binding affinity for different outputs
120
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
When we mutated and stacked ligand-binding residues in the LRR domain of LjSGN3 back to AtSGN3 versions, we found a gradual complementation of the CS formation restored, supporting a model where strong binding of ligands is necessary for CS, whereas low-affinity binding may be enough for N-signaling
110
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
We went into mechanism and asked how LjSGN3 is able to complement only N-signaling. We changed the different parts, and found that the specificity sits in the LRR-domain, suggesting that binding of CIF peptides is likely involved, and that the LjSGN3 may not work well with AtCIF ligands.

Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
thanks to Martin Stegmann (sorry, i couldnt find handle) and his group, we got a 12x mutant of CEP peptides and a CEPR double KO, which both showed similar lack of N responses upon N treatment in soil. Without displaying CS phenotypes.
120
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
with @grandpahiro.bsky.social we did some awesome phosphoproteomics and found that the differences on CAS strongly overlapped with canonical N-signaling which goes through CEP peptides and CEPR receptors. MAINLY ON SOIL.
120
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
Since we only saw this on soil, we used this uncoupling to ask, which genes are controlled by barrier formation and which are actually signaling? We compared CAS and agar plates, and found HUGE differences in responses when you put plants in actual soil..
110
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
When we grew these plants under Cologne Agricultural Soil, (CAS), which is very low on N, we found that sgn3-3 showed no N-responses in the shoots, which could be complemented by both versions of SGN3 - not only showing a connection of SGN3 to N-signaling, but uncoupling this from CS formation!!
120
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
When we complemented Arabidopsis sgn1-2 and sgn3-3 mutants with Lotus versions, we found only LjSGN1 to be able to complement CS formation defects in the respective mutant. SGN3 was not.. but, we were about to have our minds blown..
100
Tonni Andersen @tonnigrubeandersen.bsky.social · 07/07/2026
Firstly, we found that in contrast to our good old friend, Arabidopsis thaliana, the cool new root barrier model, Lotus japonicus, shows divergence in how it regulates its barriers. Here the surveillance system - mastered by the SGN3 receptor - is rewired to be part of the CS network
