TCR specificity prediction is advancing rapidly. The central question is not only which TCR sequence recognizes an antigen, but also which TCR pairings make recognition possible & where is the binding boundary? Our new paper tackles these questions. 🧵 www.science.org/doi/10.1126/...

Uneven TCR chain pairing constraints govern epitope recognition
Combinatorial pairing of independently recombined T cell receptor (TCR) α- and β-chains is central to diversifying the TCR repertoire. Although sequence motifs in one chain correlate with epitope reco...

Our paper is out! It turns out that finding the right TCR partner is a bit like matching socks🧦: almost every pair fits, but only a small fraction make the right match for antigen recognition. If you don’t feel like diving into the full paper, there’s also a one-page summary!

How much of the TCR repertoire can we make sense of? Can your TCR and GEX data be put in the context of other conditions/tissues? How many varieties of T cells are in the repertoire zoo? All these questions (& more) addressed in our latest preprint: MetaCoNGA www.biorxiv.org/content/10.1... . a 🧵

Diverse modes of T cell receptor sequence convergence define unique functional and cellular phenotypes
Single-cell techniques allow concurrent study of gene activity and T cell receptor (TCR) sequences, identifying connections between TCR structure and cell traits. Expanding on our CoNGA software, we p...

Integrating single-cell RNA and T cell/B cell receptor sequencing with mass cytometry reveals dynamic trajectories of human peripheral immune cells from birth to old age - Nature Immunology
In this Resource, authors profile peripheral immune cells from a total of 220 healthy volunteers from birth to over 90 years. This revealed that T cells were most affected by aging with divergent agin...

Inspired by @rodrahimi.bsky.social 's noble efforts linking immunologists together over here, let's try and get more specifically TCR/BCR/AIRR/repertoire chat going with this starter park go.bsky.app/Gi2dcGd Drop me a beep to get added or suggest others!
