Stefano Giandomenico @sgiando.bsky.social · 30/09/2026Correlation analysis of synaptosomes from our organoids and those from mouse cortex, prepped in the same way, shows that neural organoids recapitulate in vivo synaptic proteome enrichment patterns. 5/5 130
Stefano Giandomenico @sgiando.bsky.social · 30/09/2026To systematically characterize the protein composition of organoid synapses, we fluorescently labeled and purified synaptosomes by fluorescence-activated synaptosome sorting (FASS) before mass spectrometry. 4/5 121
Stefano Giandomenico @sgiando.bsky.social · 30/09/2026We performed proteomics across the differentiation protocol (>7,000 detected proteins per stage). Synaptic proteins ramp up sharply after day 11. 3/5 130
Stefano Giandomenico @sgiando.bsky.social · 30/09/2026We differentiated mESCs into mature forebrain organoids. They express excitatory and inhibitory synapse markers (Vglut1, Vgat, Psd95, Gephyrin) and show spontaneous calcium activity that TTX blocks, indicating action-potential-dependent network activity. 2/5 120
Stefano Giandomenico @sgiando.bsky.social · 30/09/2026Can a brain organoid grown in a dish build synapses that resemble the real thing at the molecular level? 🧠 We grew mouse forebrain organoids for 35 days and compared their synaptic proteome to that of the adult mouse cortex 🧪 www.biorxiv.org/content/10.6... 🧵 1/5 12312
