This is the equivalent of the target ID discussions where you just start stacking things on top of each other kind of as a joke but they did it.

Inspired by a conversation with @zenbrainest.bsky.social on the most important things in drug discovery. Had to beg to even get a 3rd place spot for us!
3rd place meme where a person is shown celebrating wildly in the first five panels, biting a gold medal, kissing someone, popping a bottle of champagne, and the last panel shows they are in third place

First place is chemistry, second place is chemistry, and third place is pharmacology

How it started (first three) vs how it's going (pic 4). Run your controls and hire a pharmacologist!
A screenshot of a press release header dated 19 December 2024, announcing "Certa Therapeutics Expands GPR68 Platform through Acquisition of OccuRx." Below the headline, text reads, "Strengthened Pipeline Targets Multiple Fibrotic Diseases." The body text states the acquisition is to develop precision therapies for patients with fibrotic diseases, targeting GPR68 as a "master switch of fibrosis." The Certa Therapeutics logo is visible at the top left.A screenshot of text describing recent progress. It highlights that the authors of an article focus on "Asengeprast," Certa Therapeutics' "first-in-class GPR68 proton-sensing antagonist," which has recently "completed Phase IIa clinical trials for Systemic Sclerosis (SSc), also known as scleroderma." The text emphasizes this reinforces the potential of targeting GPR68 for "fibrotic and inflammatory diseases."A photograph of Professor Darren Kelly, smiling, wearing a blue patterned collared shirt and a grey blazer. The text below describes him as the scientist who led the team that developed the GPR68 small molecule inhibitors. It notes he is currently the "CEO and Managing Director of Certa Therapeutics," a company with a portfolio of specific molecules that target GPR68.A screenshot of a withdrawn scientific preprint titled, "ASENGEPRAST: A Novel G-Protein Coupled Receptor 68 Antagonist, Reverses Clinically Relevant Inflammatory and Fibrotic Pathways in Chronic Kidney Disease." The title is clearly marked with "WITHDRAWN:". A withdrawal statement at the bottom explains the authors have withdrawn the manuscript because "key mechanistic conclusions are no longer supported by more recent experiments relating to the antagonistic effect of asengeprast (FT011) of GPR68 signaling." The DOI and a long list of authors are also visible.

Why is deformation microscopy desirable? Amino acid scale distances are relevant because of the physical properties of the individual atoms interacting with others including waters etc. in space. This is like saying "we took kublah Kahn and exploded it on the football field look how long it is!"

Very interesting but the gamma subunits have their own trafficking behaviour and halo tag is 3-5x the size of the protein itself. Hardly an innocuous alternative to tagging the g alpha.

Is your golden retriever friendly because of its breed? Genetics says: not really. A citizen science study of 18,000+ dogs found breed explains only 9% of behavioral variation in individual dogs. Personality predates modern breeds by thousands of years. #2026MMM www.science.org/doi/10.1126/...
"Effect of breed on behavior.
(A) Biddability is among eight behavioral factors defined from surveys. SE, standard error. (B) Dogs in some breeds tend to score unusually high or low for this factor compared with dogs overall. (C and D) Border collies score lower on average for biddability (vertical line at median) but vary widely (C), including genetically confirmed border collies (D). (E) In mixed-breed dogs, border collie ancestry has a small genetic effect on biddability. [Photo credits: K. Wirka (Sprocket); M. Wisniewski (Caboose); B. Hoadley (Molly); M. Logsdon (Hunter); A. A purple rectangular sign featuring a whimsical space theme. At the top center is an illustration of a green and dark-grey striped planet with a ring around it, similar to Saturn, surrounded by small white four-pointed stars. Below the planet, bold white sans-serif text asks a series of imaginative questions:
"What if... dinosaurs lived on another planet? Or dinosaurs and astronauts traveled to space together? Or there’s a whole different galaxy that’s run by dinosaurs and aliens?"
