7B: Consult with your mentors, show them notes, emails, your impression. Ask around. Don't shy away from asking bold uncomfortable questions.

7A: Write your impressions and conclusions after each meeting if possible. Do NOT let considerable time pass between meetings/interviews and note-taking. Summarize interview notes in a table and pick what fits you best. Just as with the microbiome, include your gut feeling as part of the equation.

7-Take notes: as with every step in the research process, you can't only judge by memory or vibe. Take really good notes, not of what you're told, but cross-reference what people tell you. Every person has a different story based on their priorities and personality.

6B: Talk to existing lab members, including ones you are not assigned to meet with. Schedule a zoom call or coffee later if meeting on the interview days are not possible. 6C: Visit Core facilities 6D: Email and talk to alumni. Are those scientists where you want to be?

6A: Visit the place in person for more than a day (if possible). Talk to the prospective postdoc advisor MORE than once, discuss your research interests, get a feel on whether these projects are possible in their lab or whether you'll be assigned a project.

6-Got an interview or more? Now is the time to do a thorough job of testing whether this is a common fit. These are people you will be working with for years so you have to feel comfortable working in this settings, make sure your peers and advisor are what you need.

5-Cold email PIs/labs you're interested in: 5A: Include in the email why specifically you want to do a postdoc in their lab. 5B:If you see them at conferences, reach out. 5C: If your PhD advisor can or will help, ask them.

4-Important: Very meticulously screen for good mentors. Not only great scientists, not only successful academics/PIs, but great mentors. People who would have your back and help you grow as a scientist and person.

2-Academia? Draft an area of investigation (question) or more you sense will keep you interested and the field moving forward for years, preferably decades. 3-Apply to labs studying overlapping, or similar questions or could allow you to develop the skills and knowledge needed to start your lab.

I'm surprised (and fascinated) at how nobody knew how mRNA vaccines prime CD8 T cells, despite many scientists raving about them. To my limited knowledge, this is the only example where MHC cross-dressing is a non-redundant priming pathway.

To be completely honest, this PD-1 axis gets more confusing with time. I'm not hinting that the numerous studies are contradictory, but the nuances of expression + activity and effects of targeting are increasingly complex.

Unrelated request: Would be nice if someone figured out what maintains Tcf7 expression and activity in CD8 stemlike (± mem T cells). It was argued in a recent conference that it's Wnt signaling but I can't see how in the absence of Fzd expression in T cells.

They're memory precursors. When antigen goes away, most clones contract and these cells make up the central memory (Tcm). In chronic infection, they act as transient amplifiers differentiating into terminal eff and new precursors.
