By pressurising enzyme crystals with gas and collecting X-ray data, researchers have mapped the routes CO₂ takes as it travels through the protein, revealing a 'waiting room' where substrate molecules queue up just outside the catalytic centre before being processed. www.esrf.fr/home/news/sp...

This study serves as the commissioning of the on-line UV-vis abs. spectroscopy setup available at BM07-FIP2 by the group of antoineroyant.bsky.social. With the upcoming twin paper on X-ray induced effects in buffers with Jack Stubbs and Ivo Tews, it drove the development of the icOS toolbox.

We believe this should be kept in mind when recording MSOX (mutliple sequential diffraction datasets of redox enzymes which reaction is driven by X-ray-borne free electrons), or when investigating potential SRD at RT/in structures of cryo-trapped reaction intermediates.

As a take-home message, X-ray induced features in electron-density maps cluster around the metal-centres themselves and their immediate coordination environnements at 100K, while they propagated wider in the metal-centre pocket at RT.

SRD can also be observed in CuNIR datasets at RT, albeit only at higher doses. As a consequence, we could record the first-ever RT structure (14.9 kGy) of a CuNIR with a pentacoordinated active-site Cu and two full occupancy solvent ligands, beautifully refined by Samuel Rose.

As an extension of the results presented in Gotthard et al. 2029, we find that for the most X-ray-sensitive of proteins, such as the haem-containing myoglobin, specific-radiation damage (SRD) can in fact be observed in electron-density maps at room-temperature, even with modest dose budgets.

It also evidenced that temperature altered the electron-transfer kinetics between Type 1 copper (reduced by X-ray exposure) and Type 2 copper (active site of CuNIR). Diffraction confirmed that in both cases, this is a consequence of the increased rigidity of metal centre environments at 100K.

We find that temperature is a defining factor in the response to X-ray-induced reduction of both Mb and CuNIR's metal centres. UV-vis spectroscopy identified different species of the haem iron induced by X-ray exposure in Mb (hexavalent ferrous at 100K vs pentavalent ferrous at RT).

I'm proud to present this first article from my PostDoc: We probed the different responses of metalloproteins (myoglobin, Mb and a copper-containing Nitrite Reductase, CuNIR) to reduction of their metal centres by X-rays at two temperatures (100K, 392K-RT) with UV-vis absorption and diffraction.
