A project from my postdoc in the McManus lab @mcmanuslab.bsky.social at UCSF @ucsfdc.bsky.social is out today 🎉 For years, pooled CRISPR screens have operated under a central assumption: one perturbation per cell. We challenged that assumption. 🤯 www.nature.com/articles/s41592-026-03095-w

Multiplexed perturbation enables scalable pooled screens - Nature Methods
This paper shows that high MOI sgRNA multiplexing maintains CRISPRi screen performance while enabling reduced cell numbers, challenging conventional reliance on low-MOI infections.

Small side-project from my postdoctoral work with @hitenmadhani.bsky.social. We found that blasticidin S can select for Cryptococcus transformants, and made CRISPR-compatible plasmids for other existing markers. Now 7 dominant genetic selection methods in total! journals.asm.org/doi/10.1128/...

Our small but important technical advance for conditional mutation in Cryptococcus neoformans. Combined with some other technical advances (like 100% efficient HDR), this makes working on this yeast as easy as working on S. cerevisiae. tinyurl.com/yzhjr45u
