Excited to share our latest preprint! Oftentimes, interesting stories start as a puzzling observation in the lab, and that was definitely the case here. We combined kinetic analysis, mass spectrometry, and computational modeling to solve this mystery! Grateful for the collaboration!

🚨New preprint alert! What a difference one C-F bond can make!🚨 Fluoroacetate dehalogenases (FAcDs) are remarkably good at breaking C-F bonds. But add just ONE more C-F bond to the substrate and suddenly the enzyme is inhibited. So, what’s going on? (1/4) www.biorxiv.org/content/10.6...

Substituting a conserved histidine alleviates difluoroacetate-dependent isoaspartate formation in a fluoroacetate dehalogenase
Organofluorides are ubiquitous in modern society, but their persistence poses significant environmental and health risks. Fluoroacetate dehalogenases (FAcDs) are promising candidates for the bioremedi...

Engineering Fluoroacetate Dehalogenase by Growth‐Based Selections on Non‐Natural Organofluorides
We present a high-throughput selection system to engineer fluoroacetate dehalogenases (FAcDs). By challenging E. coli populations that produce diverse FAcD libraries to grow on non-natural organofluo....

Preprint alert! With #PFAS contamination becoming an environmental crisis, we dared to engineer natural defluorinases towards degradation of (un)natural organofluorides. How? Check out our robust growth-based selection platform here: www.biorxiv.org/content/10.1... thanks to @mayerlab.bsky.social !

Two weeks ago, we had a great group outing, including a city tour through Utrecht, a Mario Kart Wii tournament, and some Rock Band Wii fun! I am truly privileged to work with such a group of talented, dedicated, and smart individuals.
