Has anyone figured out how these other non-motile bacteria get to the kidneys? As far as I have found there's nothing in Kleb.

We think this study is interesting for two primary reasons. 1. UTIs caused by Klebsiella are more likely to be pyelonephritis at the time of diagnosis 2. Klebsiella is non-motile - so how it ascends to the kidneys is intriquing.

- Despite the genetic and phenotypic diversity of K. pneumoniae UTI isolates, all strains colonized the kidneys more efficiently than bladders in a murine UTI model.

Laura Mike @lauraamike.bsky.social · 14/10/2026
- Capsule abundance and human serum resistance were significantly higher after growth in human urine; however, mucoidy, hemagglutination, and bladder cell association were not impacted by growth condition.

Highlights: - We first compared the genomic and fitness properties of 25 clinical Klebsiella UTI isolates after culturing the bacteria in LB or human urine.

Our new research article is now available! We examined the phenotypic characteristics of K. pneumoniae UTI isolates cultured in human urine vs LB. Despite diverse behaviors, these strains consistently cause pyelonephritis in a mouse UTI model. Full story here: journals.asm.org/doi/10.1128/...

In terms of impact on pathogenesis, we propose that this dynamic regulation of mucoidy by integrating multiple & opposing environmental cues is critical for Kp to efficiently colonize the gut.

Using a genome-wide screen, we found that the global regulator, cAMP-CRP is central to this sugar-driven suppression. However, cAMP-CRP doesn’t act directly on mucoidy regulator, RmpD. Instead, it requires an intermediate regulator and identifying it is ongoing in the Mike Lab.

Surprisingly, although we’ve previously reported that metabolism plays a role in distinctly regulating mucoidy and capsule, here, sugar catabolism was dispensable. Import alone is sufficient to dictate the mucoidy state of hypervirulent Kp.

Laura Mike @lauraamike.bsky.social · 26/08/2026
We screened 11 monosaccharides for their effect on mucoidy and observed that that sugars generally suppress Kp mucoidy in a concentration-dependent manner.

Last year, we reported that arginine stimulates mucoidy in hypervirulent Klebsiella pneumoniae. Now we show an opposing cue, sugars! Here, we report that sugar import suppresses mucoidy. Full story in @natcomms.nature.com: www.nature.com/articles/s41... #microsky #klebclub See 🧵 for a summary:

Thanks Roby! This one was a labor of love - it's took awhile to figure out how to put the paper together. What we find very interesting is that despite many isolate to isolate differences and changing phenotypes in urine, all the strains colonize kidneys very well.

Clinical relevance: K2 is a common serotype in neonatal sepsis and community-acquired (hvKp) and hospital-acquired (cKp) infections. Capsule property variations within this serotype affects pathogenesis.
000
Laura Mike @lauraamike.bsky.social · 11/02/2026
Why it matters: Controlling for K2 serotype revealed that mucoidy, siderophores and macrophage evasion, NOT capsule abundance or complement resistance, distinguish hvKp bloodstream fitness. This suggests cell-mediated immunity (not just complement) is critical for controlling Kp bacteremia.

Laura Mike @lauraamike.bsky.social · 11/02/2026
We found: ✅ hvKp more mucoid (uniform CPS chains) ✅ hvKp better evade macrophage association ✅ cKp have higher C3b deposition (but similar serum survival!) ✅ hvKp achieve higher spleen/liver burdens in vivo ✅ NO difference in capsule abundance, growth kinetics, or blood survival

Divergent roles for complement components C3 and C4 in controlling Klebsiella pneumoniae gut colonization and systemic dissemination | mBio
Klebsiella pneumoniae, a major public health threat, resists antibiotics and can spread from the gut to the bloodstream, causing severe infections. Our study reveals how the immune system uses complement proteins C3 and C4 to block this spread. C3 limits bacterial growth in the gut through two potential mechanisms: (i) coating K. pneumoniae with fragments that signal bacteria-eating phagocytic cells to destroy it and (ii) recruiting more phagocytes into the gut. C3 also helps clear bacteria that escape into the blood. However, when antibiotic-resistant strains overgrow, C3 alone is insufficient. In these cases, C4 becomes critical, likely by enhancing C3’s ability to tag bacteria for elimination. This two-layered defense highlights new immune pathways that could be targeted to prevent bloodstream infections, especially in vulnerable patients or those colonized with drug-resistant bacteria. These insights open doors to innovative strategies against life-threatening Klebsiella infections.
