Harnessing the murine inner cell mass mechanical environment enhances derivation of in vitro nascent primitive endoderm precursor cells Read this Article from our special issue #DevSIextracellular by Elena Corujo-Simon, Jennifer Nichols, @kevinchalut.bsky.social & co. doi.org/10.1242/dev....
Figure from Elena Corujo-Simon et al.

NPRE FGF+ cells aggregate with mESCs and contribute to host PRE in mouse blastocysts. (A) Representative confocal images of ICM aggregates made of 30 NPRE cells and 20 mESCs 8, 24 and 48 h after initial seeding. Pdgfrα H2B-GFP/+ FGF+ NPRE cells (green) and mESCs immunostained for OCT4 (magenta) are shown with nuclei stained with Hoechst. (B) Chimera injection diagram. Five NPRE cells were injected into host blastocysts at E3.5. Embryos were allowed to develop until E4.5 prior to fixation and immunostaining. (C) Representative confocal images of injected NPRE cells contributing to the host PRE in the mouse blastocyst (top) or attaching and contributing to the TE (bottom).

It was really fun to work on this collaboration with @storerlab.bsky.social and Byron - I think we are really starting to see that ECM is more than just a scaffolding. It's really driving tissue function in important ways, like in this application of tissue repair.

🚨 New Preprint from the Paluch & Chalut Labs 🚨 Ana Raffaelli et al show that the stiffness of substrate (in vitro) / basal membrane (in vivo) determine how cells interpret biochemical signals (BMP4) and which fates they acquire #mechanobiology #hESCs #substratemechanics doi.org/10.64898/202...

Excited to share this new work from Ana Raffaelli, in collaboration with Ewa Paluch's lab and involving several of my colleagues at Cyclana Bio. In this paper, we show that basement membrane mechanics instructs developmental signalling and fate patterning. www.biorxiv.org/content/10.6...

Excited to share our work on epithelial multilayering - identifying why stem cell stay in the basal layer and how and why differentiating cells move up. Great collab with @manningresearch.bsky.social and Niessen labs! Check out preprint and great summary below www.biorxiv.org/content/10.6...

Very excited that the lab's first paper is out! We showed that in stem cells, Aurora B controls abscission dynamics by regulating microtubules stability and MCAK recruitment. Very happy with this streamlined version of our 2024 preprint. www.cell.com/cell-reports...

Aurora B controls microtubule stability to regulate abscission dynamics in stem cells
Mouse embryonic stem cells remain connected by bridges for hours after cell division. In this paper, Kodba et al. show that a high Aurora B activity at the bridge leads to microtubule stabilization. S...

We are really proud to share the first 'Pre-Print' from the Storer lab driven by the fantastic Byron Mui, and co-lead by the innovative Kevin Chalut, on tissue mechanics, ECM and regeneration. A big thanks also to our collaborators in the Franze Lab. www.biorxiv.org/content/10.1...

Hyaluronic Acid and Emergent Tissue Mechanics Orchestrate Digit Tip Regeneration
Tissue regeneration is a dynamic process requiring coordinated cell fate decisions to restore structure and function. For instance, the tips of human and rodent digits fully regrow after amputation, a...
