We are excited to share the news that the @mssocietyuk.bsky.social has renewed our funding for 5 years! We will continue our work, understanding and developing treatments for MS! @cam.ac.uk @camneuro.bsky.social @scicambridge.bsky.social @karadottirlab.bsky.social @pluchinolab.bsky.social

Focal white matter lesions drive grey matter inflammation and synapse loss - Nature
A transient grey matter microgliosis is required for remyelination, the failure of which results in a chronic neuroinflammatory state seen in neurodegenerative disorders.

This reframes how we think about neurodegeneration: white matter lesions and grey matter pathology are coupled through a regenerative plasticity program. If we can promote myelin regeneration, we may be able to prevent chronic inflammation that drives MS, Alzheimer's, and related diseases. 🧵 (6/8)

What was surprising was that this grey matter inflammation was not harmful, but essential for myelin regeneration. When we blocked it, remyelination failed. And when remyelination fails, transient inflammation turns chronic — matching the low-grade inflammation seen in neurodegeneration. 🧵 (5/8)

The effects were striking: • 60% drop in neuronal firing • 3× increase in microglial density around neuronal cell bodies • Transient loss of excitatory synapses, engulfed by microglia All ~3.5 mm away from the lesion. 🧵 (4/8)

White matter lesions appear early in MS, Alzheimer's, Parkinson's. But their functional impact on neurons has been unclear — and they've been largely dismissed as secondary to neurodegeneration. 🧵 (2/8)
