Here are the results in terms of the number of projects. The number of funded projects in FY2025 is 4014 fewer than the number for FY2024. 2/3
A grant showing the number of projects funded for FY2021 through FY2025. The number for FY2025 lagged behind and did not catch up with 4014 fewer projects funded in FY2025 compared with FY2024.

The fiscal year ends tomorrow. Here are results from NIH Reporter downloaded an hour ago. The total amount of funding committed for FY2025 at this point is 99.0% of that for FY2024. The same difference could be due to a variety of technical factors. 1/3

Oversimplified immunology is holding biomaterials back
Nature Reviews Bioengineering, Published online: 19 May 2025; doi:10.1038/s44222-025-00320-7Immune responses are complex, often defying rigid classifications. Instead of interpreting results according to reductionist categories, researchers should rely on comprehensive single-cell data to guide analysis and should remain open to unexpected immunological complexity.

Add to this, the slow pace of new and competitive renewal awards due to study section and advisory council meeting delays and actual grant terminations, the flow of funds has been reduced and has become more unpredictable. This is no effing way to run the world’s best biomedical research system.

ICYMI, NIH will be asking soon for each NIH-funded researcher to justify every draw they make (often several times a week) on their grants and every NIH officer and their boss will have to justify their agreeing on these draws. This is the death of US biomedical research by a thousand cuts

Production of site-specific antibody conjugates using metabolic glycoengineering and novel Fc glycovariants
Molecular conjugation to antibodies has emerged as a growing strategy to combine the mechanistic activities of the attached molecule with the specificity of antibodies. A variety of technologies have been applied for molecular conjugation; however, these approaches face several limitations, including disruption of antibody structure, destabilization of the antibody, and/or heterogeneous conjugation patterns. Collectively, these challenges lead to reduced yield, purity, and function of conjugated antibodies.

Excited about the therapeutic potential of branched PBAEs developed by Erick E Rocher, a phenomenal undergraduate student at Hopkins BME applying to #MDPhD programs, now out in Biomacromolecules (pubs.acs.org/doi/full/10....)
pubs.acs.org
Efficient Polymeric Nanoparticle Gene Delivery Enabled Via Tri- and Tetrafunctional Branching
Poly(β-amino ester) (PBAE) nanoparticles (NPs) show great promise for nonviral gene delivery. Recent studies suggest branched PBAEs (BPBAEs) offer advantages over linear counterparts, but the effect of polymer structure has not been well investigated across many chemical constituents. Here, a library of BPBAEs was synthesized with tri- and tetrafunctional branching. These polymers self-assemble with DNA to form highly cationic, monodisperse NPs with notably small size (∼50 nm). Optimal transfection occurred with polymer structures that featured moderate PBAE branching, enabling complete DNA encapsulation, rapid NP uptake, and robust expression at low DNA doses and polymer amounts. Optimized NPs enabled efficient DNA delivery to diverse cell types in vitro while maintaining high cellular viability, demonstrating significant improvements over a well-performing linear PBAE counterpart. BPBAEs also facilitated efficient mRNA and siRNA delivery, highlighting the versatility of these structures and demonstrating the broad utility of BPBAE NPs as vectors for nucleic acid delivery.
