In his Int J Cancer editorial, @jeffreytownsend.bsky.social cautions that a pan-cancer co-mutation map can reflect tissue. Vaeyens et al. published their data, so we re-ran their BRAF/KRAS/EGFR calls within tumour types and against chance. Textbook exclusions hold; no co-occurrence does.

The 59 co-mutation calls Vaeyens et al. flag as significant, grouped by the direction each names and re-tested on the authors' data against chance within tumour types at the study's code threshold. Class exclusivities: 7 of 8 hold, 1 cannot be settled within tumour types. Variant exclusivities: 6 of 7 hold, 1 in the same direction at p below 0.05 only. Variant-pair exclusivities: 9 of 38 hold, 29 cannot be settled within tumour types. Co-occurrences: 0 of 6 hold; 4 are consistent with chance and 2 are found together less often than chance.

BRAF V600E already guides targeted therapy; PI3K targeting remains investigational. However, trials often stratify colorectal cancers by left vs right side rather than colon versus rectum. Parsing by anatomical site may therefore reveal clinically relevant variant-specific biology. 🧪 rdcu.be/fx0fu

Colon tumors had more mismatch-repair-associated mutational signatures and a higher mutation burden than rectal tumors. However, these differences can make mutations more common without making them more advantageous. For tissue biologics, we aligned cancer-effect estimates across cohorts. 🧪
