Overall, we find that intestinal secretory progenitors respond to a second Notch signaling event by expressing meis1b to become best4+ cells, rather than enterochromaffin cells. pbx3a then works to make them spatially heterogeneous in gene expression and function.

Additionally, we improved the electron microscopy showing the morphology of basolateral projections significantly, added some additional stainings, and the entire manuscript is leaner and meaner, all thanks to comments from invested and helpful reviewers.

While under review, Ella managed to watch best4:pHluorin2 cells live in vivo while exposing them to different pHs and showed that best4+ cells are functionally heterogenous: anterior ones respond to acidic pH but not posterior by intracellularly acidifying.
More acidic solutions cause more dramatic intracellular pH changes in anterior best4+ cells, while posterior best4+ cells respond the same to solutions of different pHs.

Hard to say goodbye today to the lab’s first postdoc, but so excited to see what @abhinavsur.bsky.social will do in his new lab at U Arizona. We’re going to miss you deeply over here. Safe travels and knock em dead.

If you love Daniocell, but wish it could generate analyses specific to YOUR genes & cell types of interest, check out DaniocellDesktop - a new point-and-click app for Mac and Windows that enables reanalysis of the Daniocell data without programming: daniocell.nichd.nih.gov/desktop/ (1/4)

Thus, we made single to quadruple crispants of UPR-associated TFs to identify which were required for the UPR in each cell type. We also mis-expressed each UPR TF across all tissues and to identify which transcriptional programs they were sufficient to activate. (7/)
Notochord and hatching gland cargoes are not correctly secreted after unfolded protein response transcription factors are removed.

Out now in Dev Cell! scRNAseq trajectories describe the cascade of gene expression as cells differentiate, but turning them into a clear understanding of the underlying biology of a cell type's development remains challenging. authors.elsevier.com/c/1k9qj5Sx5g... (1/)

So excited to catch up with you wonderful dev biologists at #SDB2022! Hope to see you at @abhinav_sur's talk about our new zebrafish scRNAseq data, shared gene modules, and uncharacterized cell types. And so much more he can't fit in the talk - chat with him after!

Very bittersweet see Eric and Jeremy off to their next steps this week after two awesome years as post-bacs in the lab. So proud of everything they accomplished and can't wait to see what they do next. Especially looking forward to finding out which lab Eric joins @WashUDevBio.

Huge congratulations to @abhinav_sur on winning a poster award at #IZFC2022. And just so we’re clear, it was an award for scientific content, not its incredible, record setting size. #proudpi

This weekend, celebrated my PhD mentor Pat O’Farrell’s 70th birthday with most of the rest of the former members of the O’Farrell lab. We were all thankful to be shaped by his curiosity, passion, rigor, and generosity. He gives us all a high bar to live up to.
