🧬 Checkpoint mutants undergo gradual degradation of nascent DNA at replication forks, which is prevented in the absence of Rad51, Mus81, Rad5 and Exo1 🧪 Heterologous expression of human PrimPol or overexpression of Mre11 can counteract the aberrant processing of replication forks

📄 We identified Rad51, Mus81 and Rad5 to be involved in the irreversible replication fork arrest observed in yeast checkpoint mutants upon DNA damage, despite they are also required for normal fork progression or restart once the damage is removed

S-phase checkpoint protects from aberrant replication fork processing and degradation
Abstract. Replication stress, a hallmark of cancer cells, is detected by checkpoint mechanisms that trigger a range of cellular responses. Among these, the

Mechanism of DNA replication fork breakage and PARP1 hyperactivation during replication catastrophe
Upon ATR inhibition, APOBEC3B targets unprotected single-stranded DNA at replication forks, leading to fork breakage.

Glad to see our PACT preprint published in @naturecomms.bsky.social! "Cooperative role of PACT and ADAR1 in preventing aberrant PKR activation by self-derived double-stranded RNA" Another step forward in understanding innate immunity. @buissonlab.bsky.social 🔗 www.nature.com/articles/s41...
