Very excited to share our newest preprint, led by myself and Biostats PhD Student Jianing Yao, where we generate a three dimensional reconstruction of the human NAc to understand how cell type heterogeneity and topography change across the anterior-posterior axis. www.biorxiv.org/content/10.6...

Excited to share work with cryostat wizard @heenadivecha.bsky.social looking at gene expression in my favorite brain region (LC) in adult postmortem brain! (Long URL to spare the trouble of identifying cars, buses, beds in 5x5 pixel imgs). (1/x) link.springer.com/epdf/10.1007...

I am very excited to see this work out and so proud of @mictott.bsky.social & @svitlanabach.bsky.social! Study is a great example of moving beyond generating spatial transcriptomics data simply to generate molecular maps, to using the data to answer longstanding questions about human 🧠 organization.

👀 - me watching project channels on Slack on Fridays for #FluorescenceFriday contributions... literally fresh off the scope from JHU undergrad Mahima Sanghera, @ishbeldelrosario.bsky.social and @svitlanabach.bsky.social some dorsal raphe in human 🧠 labeling 5HT neurons - 🟣TPH2 and 🟡 SLC6A4
Fluorescence microscopy image of section of human brain at the level of the dorsal raphe nucleus using RNAscope for anatomical validation of the presence of serotonin neurons, here with overlapping signal of yellow and purple.

For #FluorescentFriday, some more D1-island RNAscope from @ishbeldelrosario.bsky.social. These cells are so weird in all the best ways. Excited to share this data with everyone soon 😎🏝️

Since #FluorescenceFriday usually gets the spotlight, taking today to give brightfield microscopy some love with this really beautiful image of a Nissl stain in the dorsal raphe nucleus of the human 🧠. Image credits to @ishbeldelrosario.bsky.social @svitlanabach.bsky.social 🔬👩‍🔬🧪

For #FluorescenceFriday and in celebration of a paper acceptance - super excited to get this one out to the community soon 🤩, cluster of "D1 islands" in the nucleus accumbens of the human 🧠 (🟢 MBP; 🔴 DRD1; 🟡 RXFP1; 🟣 OPRM1). And some love for the human 🧠 for #FluorescenceFriday. I like these zoom outs where you can see a whole structure - here, striatum. Green is MBP and outlining the white matter tracts, 🟣 is DRD1 and if you zoom in (see zooms in following posts) there is 🔴 for RXFP1 marking clusters of island cells.

Early for #FluorescenceFriday, but got an early peek in a project meeting - strucure is just so incredible - zoom in of tryptophan hydroxylase 2 labeling 🟣 of serotonin neurons in the dorsal raphe of human 🧠. Experiment and image credit to @ishbeldelrosario.bsky.social @svitlanabach.bsky.social

Too excited about this image to wait for #FluorescenceFriday 🔬🤩👩‍🔬. Serotonin neurons 🟣 in the raphe nucleus along the midline of the human brainstem 🧠, flanked bilaterally by norepinephrine neurons 🔴 in locus coeruleus. Image credit to @ishbeldelrosario.bsky.social @svitlanabach.bsky.social
Microscope imaging labeling mRNA transcripts in situ of postmortem human tissue in the transverse plane of the brainstem labeling tryptophan hydroxylase 2 (TPH2) and dopamine beta hydroxylase (DBH), to label serotonin neurons in the raphe, and norepinephrine neurons in the locus coeruleus, respectively.
