How does the RAS effector AFDN bind the polarity protein SCRIB? Our Biochemical Journal paper identifies a distinctive β-hairpin interaction using AlphaFold3 combined with biophysics. Surprisingly, disrupting SCRIB-binding prevents AFDN from complexing with activated KRAS. doi.org/10.1042/BCJ2...

This was a true team effort. IRIC chemists generated complex macrocycles with the goal of targeting KRAS and my former postdocs Ryan Killoran and Changhwa Jo offered insights to how the macrocycles were targeting RAS using NMR and crystallography. This work continues! pubs.acs.org/doi/10.1021/...
