My first 3 years as a TTF we're filled with grant rejections, including an MRC NIRG that I was devastated about. It's similar for many others, just not talked about as openly as the successes. Resilience is important in research. When you get that first break you can start to build some momentum.

It may be relatively unique to paracetamol (acetaminophen) given the need for relatively large doses being converted to the toxic reactive metabolite. But it's also more obvious from the literature given how often paracetamol is used as a reference hepatotoxin and the availability of clinical data.

Ian Copple @iancopple.bsky.social · 21/11/2024
This finding highlights the limitation of using acetaminophen as a reference hepatotoxin for the evaluation of in vitro liver models. Hence, we make recommendations on the selection of reference hepatotoxins for this purpose.

Ian Copple @iancopple.bsky.social · 21/11/2024
Using the cytochrome P450 inhibitor 1-aminobenzotriazole, we show that acetaminophen toxicity in cultured human, mouse, and rat hepatocytes is not dependent on N-acetyl-p-benzoquinonimine formation, unlike the in vivo setting.

Ian Copple @iancopple.bsky.social · 21/11/2024
However, the induction of substantial cell death in these models typically requires acetaminophen concentrations (∼10 mM) far higher than blood concentrations of the drug associated with clinical hepatotoxicity (∼1 mM).

Here's the abstract: Acetaminophen is commonly used as a reference hepatotoxin to demonstrate that in vitro human liver platforms can emulate features of clinical drug-induced liver injury.

Are you developing in vitro human liver models? Great! But be careful using paracetamol (acetaminophen) as a reference hepatotoxin - see our new article in Tox Sci for the details. Led by former PhD student Lucia Livoti. #LiverSky academic.oup.com/toxsci/advan...
