We will not understand antimicrobial resistance by studying genes, cells, or populations in isolation. Our new mini-review argues for frameworks that connect these scales: From cells to populations: Multi-scale quantitative approaches to antimicrobial resistance www.sciencedirect.com/science/arti...
Figure 1 of the paper. Multi-scale quantitative approaches to antimicrobial resistance (AMR). Population-level models (left) address (a) dose optimization, (b) sequential and combination therapies exploiting collateral sensitivity, (c) community effects on drug response, and (d) global epistasis. Cellular-level models (right) describe (e) resource allocation among ribosomal, metabolic, and stress sectors and (f–h) non-linear physiological transitions producing bistability and switches between susceptible and tolerant states. Both levels combine modeling with controlled experiments; they differ in biological organization rather than method.

This work provides a structural and mechanistic explanation of how transient CS bypasses historical contingency and minimizes the risk of selecting stable resistance mutants, opening immediate avenues for the design of rational combinatorial therapies.

We previously described a rational solution (DOI: 10.1038/s41467-023-37357-4) to the lack of robustness in mutation-driven collateral sensitivity (CS). Here we explain the molecular mechanism behind transient CS to tobramycin associated with MexCD-OprJ over-production: OprM promiscuity.

In summary, Why matters?: • No microbiome is truly “pristine” • Environmental factors (like Hg pollution) are relevant selective pressures • Resistance can spread fast even in low-impact communities • Definition of "backbone" and "baseline" resistomes are key to interpret AMR
