Special thanks to Giuseppe Ianiri, whose expertise in fungal genetics and invaluable collaboration were essential to this work. His insight, support, and enthusiasm played a key role in many of the discoveries presented here.

This project would not have been possible without an outstanding team of collaborators. I am especially grateful to @lableibundgut.bsky.social for her mentorship, trust, and support throughout my PhD, and to all members of the lab for making it such a collaborative environment.

Malassezia is often viewed as an opportunistic pathogen linked to inflammatory skin diseases. Our findings reveal a beneficial side of this abundant skin commensal, showing that it can promote barrier maintenance and tissue homeostasis through evolutionarily conserved AhR signaling pathways.

Using fungal genetics, metabolomics, human epidermal models and mouse experiments, we showed that these metabolites promote skin barrier restoration and reduce inflammation. Using mutants defective in indole production, we established that this protective effect depends on fungal AhR ligands.

After several years of work, I'm happy to share that the main project from my PhD is now published in Nature Microbiology. We show that the skin commensal Malassezia produces tryptophan-derived indoles that activate AhR promoting skin barrier function and tissue homeostasis. doi.org/10.1038/s415...

The rules of immunity are different in early life! Lead author Dr. Yue Xing finds that early life DCs sense allergens and trigger inflammation directly in skin. This in-situ activation state emerges from a developing HPA and low glucocorticoids. #Naiklab @nature.com www.nature.com/articles/s41...

Malassezia is typically viewed as an opportunistic pathogen — but for the first time, we show evidence that it plays a mutualistic role in the skin.

"Ultimately, these changes lead to insufficient activation of a pathway controlled by the aryl hydrocarbon receptor (AHR)" not to be that smug guy on the Internet but this is why I keep yelling about AhR and Regulatory T cells and immune conditions liminalnest.wordpress.com/2024/05/28/b...
