GBE | In Vitro Massively Parallel Screening of Human Regulatory Elements Involved in Postcranial Skeletal Development for Differential Activity Compared to Chimpanzee

Massively parallel reporter assay (MPRA) experimental design. Regulatory elements accessible across the developing human skeleton were standardized to a length of 540 base pairs and divided into two tiles to design an MPRA library that included both the human and chimpanzee sequences for each tile. This library was then transfected into three cell lines (2 distinct cartilage, one  myeloid) to measure the relative potential of the human and chimp sequences to drive reporter gene expression (GFP – green fluorescent protein).
