New preprint: does heterochromatin phase separate? Our answer — it's not about if but when. MOCHA-FRAP across 7 cell lines and 10 proteins shows barrier compartments coexist with non-barrier ones in the same nucleus, set by protein enrichment and competition. www.cardoso-lab.org/pages/public...

New in Nucleic Acids Research! MBD2 forms liquid condensates that organize heterochromatin and control histone acetylation. Combined proteomics + AI predictions reveal how phase separation regulates chromatin states. MBD2 recruits HDACs, excluding HATs. doi.org/10.1093/nar/...

MeCP2driven chromatin organization controls nuclear stiffness during neuronal differentiation. UsingAFM, we show heterochromatin clustering increases nuclear mechanics, disrupted by Rett mutations-severity correlates with stiffness defects, not gene expression. www.nature.com/articles/s42...

A new way to watch chromatin moving in living stem cells as they become neurons and what we saw was striking. Chromatin literally slows down as cells commit to their fate. It's like watching cells physically lock in their identity by constraining their chromatin. cardoso-lab.org/pages/public...

Our new work in EMBO Reports! 🧬 H3K36me3 enriches at pericentromeric heterochromatin BEFORE replication, regulating its timing. H3K36me3 loss disrupts satellite transcription and delays replication. Forward-strand MajSat RNA controls replication timing—ncRNAs as key regulators! cardoso-lab.org
