8/8 Overall, these findings suggest that the rhythmic organization of sleep - not just its duration or architecture - may provide a new window into the biology of neurodegeneration in Alzheimer's.

7/8 The EEG findings also showed relationships with molecular markers. ISO amplitude showed a trend-level association with the Aβ42/40 ratio, while ISO bandwidth was associated with neurofilament light (NfL) and showed trends involving GFAP and memory retention.

6/8 So a disrupted sleep rhythm could potentially connect: locus coeruleus → sleep microarchitecture → glymphatic function → Alzheimer’s pathology

Brain and Sleep Research Laboratory @bsrlaboratory.bsky.social · 04/10/2026
5/8 There’s another intriguing connection: Noradrenergic activity during NREM sleep may influence blood vessels, astrocytes and cerebrospinal-fluid movement, all processes thought to support glymphatic clearance of metabolic waste, including amyloid-β.
100
Brain and Sleep Research Laboratory @bsrlaboratory.bsky.social · 04/10/2026
4/8 Why might that matter? One hypothesis is that these infraslow oscillations reflect rhythmic activity of the locus coeruleus, a brain region involved in noradrenaline signaling. The locus coeruleus is also one of the earliest regions affected by Alzheimer’s pathology.

Brain and Sleep Research Laboratory @bsrlaboratory.bsky.social · 04/10/2026
3/8 We found that the amplitude of the infraslow oscillation was significantly reduced in people with Alzheimer’s disease. Importantly, its frequency and bandwidth were preserved. So this wasn’t a general disruption of the rhythm. It was a selective weakening of its strength.
100
Brain and Sleep Research Laboratory @bsrlaboratory.bsky.social · 04/10/2026
2/8 We asked a simple question: Is this infraslow organization of sleep spindles altered in Alzheimer’s disease? We analyzed multiple overnight EEG recordings from 30 older adults: → 10 with Alzheimer’s disease → 20 cognitively unimpaired controls

1/8 During NREM sleep, the brain produces bursts of activity called sleep spindles. These are important for memory and reflect coordinated communication across brain networks. But spindle activity isn’t random. It waxes and wanes in a much slower rhythm: an infraslow oscillation

🧠 What if the connection between sleep and Alzheimer’s disease is partly hidden in a rhythm of the brain that unfolds much more slowly than typical brain waves? Our study in Alzheimer’s & Dementia points to an intriguing candidate: infraslow oscillations of sleep spindle activity.

Are infraslow oscillations the missing link between sleep and Alzheimer's?
INTRODUCTION Locus coeruleus (LC) and glymphatic dysfunction have been linked both to Alzheimer's disease (AD) and, recently, to infraslow oscillation (ISO) in sleep spindle (sigma) activity (ISO). ...
