Would you expect that Bacteria use an archaellum for swimming? We didn't, but we found that some Chloroflexota do! Find the story here: www.biorxiv.org/content/10.1... A little thread below 1/n

Horizontal gene transfer of the functional archaellum machinery to Bacteria
Motility in Archaea is driven by a nanomachinery called the archaellum. So far, archaella have been exclusively described for the archaeal domain; however, a recent study reported the presence of arch...

The story is finally out!!! Here, we showed multiple functions of the VapBC4 toxin-antitoxin of Sulfolobus acidocaldarius under stress. Thanks to @archaellum.bsky.social for an excellent collaboration. journals.asm.org/doi/10.1128/...

Role of VapBC4 toxin-antitoxin system of Sulfolobus acidocaldarius in heat stress adaptation | mBio
This research enhances our knowledge of toxin-antitoxin (TA) systems in archaea, specifically in the thermoacidophilic archaeon Sulfolobus acidocaldarius. TA systems are widespread in both bacterial and archaeal genomes, indicating their evolutionary ...

In parallel, Yulong Shen et al also found components of these machineries in S islandicus and you can also read their paper in BioRXiv Really nice how they used a CRISPR plasmid to induce DNA damage www.biorxiv.org/content/10.1...

Identification of novel components of the Ced and Ups systems in Saccharolobus islandicus REY15A
In the cells of Sulfolobales, the Ced (Crenarchaeal system for exchange of DNA) and Ups (UV- inducible pili of Sulfolobus) systems are highly induced by DNA damages and function in chromosomal DNA exc...

With these 2 new components we expand our knowledge of the response of Sulfolobus to dsDNA damage but there are still several questions remaining (and probably components missing), specially on the intriguing Ced system Read more about it in our preprint www.biorxiv.org/content/10.1...

While UpsC has a characteristic pili like folding, CedD is homolog to VirD4, a protein that forms part of the conjugation machinery in bacteria The Ced system is homologous to the conjugational machinery but acts in a different direction, by importing DNA

After the cells are in contact, the Ced system acts as an importer to bring DNA from the neighboring cell inside. This mechanism is independent of the Ups pili. Deletion of saci_0667 now cedD, avoids import of DNA, as it is the case for other deletion strains of Ced-sys

The Ups pili is in charge of creating cell-cell contact in a species-specific manner. When saci_1302, now upsC, is deleted, cells cannot form aggregates anymore

After UV light or damage to the dsDNA, Sulfolobus cells repair these damage by exchanging DNA between them and performing homologous recombination. Along side the previously known Ups and Ced genes, other genes are also up regulated as response

Our latest pre print is out! We (@marleenvw.bsky.social @sshamphavi.bsky.social at @archaellum.bsky.social lab) studied up regulated genes in UV-stress and found two new components of the Ups pili and the fascinating Ced system in Sulfolobales #ArchaeaSky #MicroSky www.biorxiv.org/content/10.1...

New components of the community based DNA-repair mechanism in Sulfolobales
After exposure to UV light, Sulfolobus acidocaldarius cells aggregate in a species-specific manner to exchange DNA and repair double-strand breaks via homologous recombination. The formation of cell-c...
