Hi! I’m @vitoryang.bsky.social, a PhD candidate studying how cells migrate collectively during development. My work focuses on cytoskeleton dynamics and Rho GTPase signalling, using Drosophila border cells as a genetically tractable in vivo model.

The model of cell invasion and migration that I have been studying over the course of 7 years in finally featured in Journal of Cell Biology. 🧬 I couldn’t be any more happy to have my work published there as the whole process was streamlined and very clear! My experience was highly pleasant 🥳

I am so happy to share that my previous preprint is officially out as a Report in JCB. 🩵 Hop in to find out more about how cells move as an invasive cluster 🧬

All in all, RhoGAP15B locally suppresses RhoA–Myosin II at the leading edge, creating a relaxed cortical zone that stabilises protrusions while possibly helping to dictate the cycles of contractility/extension, thereby setting the pace for migration.

RhoA activity (measured with a FRET sensor) is increased with RhoGAP15B RNAi. While there is normal rear–front asymmetry in migrating border cell clusters, the loss of RhoGAP15B leads to elevated, spatially uniform RhoA activity, consistent with excessive cortical Myosin-driven contractility.

RhoGAP15B:GFP shows predominantly cortical localisation, with enrichment at the outer cortex. Within the leader cell, RhoGAP15B concentrates at the leading edge and is largely excluded from the nucleus and cytoplasm, consistent with a role in locally modulating RhoA–Myosin II at the migration front.

RhoGAP15B RNAi collapses leader protrusions, while RhoGAP15B OE stabilises and extends them, underscoring tight control protrusion dynamics.

How do migrating cell groups balance force and flexibility? Our new preprint shows that RhoGAP15B downregulates RhoA–Myosin activity to stabilise protrusions and enable efficient collective migration in vivo. Happy New Year! www.biorxiv.org/content/10.6...

Porto is a beautiful during spring (it is all year around but I may be biased)! And is hosting the conference that you don't want to miss on #adhesion regulation in #cancer. A great lineup of speakers from basic to clinical research and also many talk opportunities. Check it out and join us!
