Academia relies on good faith. It cannot be done otherwise. This is precisely why plagiarists, people who fake data, people who deliberately ignore counter-evidence, and fascists need to be driven out. Full cordon sanitaire against this crap.

What does accelerated biological aging actually mean for children? Can we measure it with algorithms only trained in adults? And do different biomarkers of aging measure the same thing? Find out at my #SOBP2026 talk tomorrow afternoon 🧬🧠

13/13 Huge thanks to everyone involved – especially the Kids2Health participants! This work is only possible because way more children than expected were brave enough to give blood samples! If you have questions about the data, or want to discuss implications of the results, please get in contact 🧬

12/13 We need more epigenetic measures for pediatric populations – ideally valid across tissues. You can use our epigenome-wide correlation database to train such measures, e.g. by only selecting CpGs with high cross-tissue generalizability. Download the data here: doi.org/10.5281/zeno...

11/13 What can we conclude from this? An ideal epigenetic measure should correlate highly and perform equally (scenario 1) in blood and saliva. Only 1/22 measures met both criteria (most measures met just one). So, be cautious when applying epigenetic measures outside their training age and tissue!

10/13 What does this mean for previous EWAS results? We downloaded significant EWAS hits and calculated their average cross-tissue correlation based on our data. Many health-relevant traits (e.g. immune & metabolic) showed small correlations, implying high tissue-specificity of EWAS hits.

9/13 Moving from aggregated measures to epigenome-wide analyses, we estimated within-person blood-saliva correlations for 815,069 CpGs. Correlations were low for most CpGs (mean rho=0.15). Only a small subset (2-3%) of CpGs was variable and highly correlated – maybe useful for training new measures.

8/13 Next question: are previously reported effects of early-life adversity and SES consistent across tissues? Yes – effect directions were mostly the same in blood and saliva. We found a consistent signature of ELA/SES on elevated epigenetic CRP and lower epigenetic-g (trained on cognition).

Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
7/13 Expanding previous work by Abner Apsley & @sepidehzarandooz.bsky.social, we examined within-person correlations of epigenetic measures between blood and saliva. 6/22 measures correlated strongly between tissues. Concerningly, age acceleration estimates were moderately correlated at best.
110
Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
6/13 Same for epigenetic scores: in scenario 1 (including 3 scores), epigenetic CRP was not associated with measured CRP in saliva, but behaved similarly to blood after cell type correction. In scenario 2, salivary DNAmTL did not correlate with measured telomere length no matter what.
100
Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
5/13 In scenario 1, associations of GrimAge2 and chronological age differed in blood and saliva, but these differences are attenuated with cell type correction. 6 clocks fall into scenario 1. In scenario 2, PedBE clearly performed better in saliva than blood regardless of cell type correction.
100
Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
4/13 We first checked: how are epigenetic and chronological age correlated, i.e. how well do epigenetic clocks perform, and does this differ between tissues? Results can be clustered into 2 scenarios.
100
Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
3/13 But can we generalize across age groups and tissues? We investigate this in the Kids2Health cohort, collected at #Charite Berlin. We calculated 22 common epigenetic measures in matching blood and saliva methylation (EPICv2) from 291 children aged 3-12, 56% exposed to early-life adversity.

Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
2/13 Epigenetic clocks and scores are investigated in children as biomarkers for health outcomes following early-life exposures. The problem is: most epigenetic measures are trained in adult blood, while saliva samples are more readily available in pediatric cohorts.

Vera Karlbauer @vera-karlbauer.bsky.social · 17/12/2025
1/13 Can epigenetic clocks and scores trained in adult blood be applied to blood and saliva in children? We systematically explore this in our new pre-print in a cohort with 291 matched pediatric blood and saliva samples @binderlabmpip.bsky.social 🩸💧🧬🧒🧵 doi.org/10.64898/202...
