Among many additions since the preprint: a divergent DFG-like (DPV) motif that likely locks T7K in the ON mode, and the discovery of T7K homologues in jumbophages.

This function was missed for ~50y b/c T7 biology was mainly explored in lab strains w/ few defenses. Our infection assays in natural isolates show strong phenotypes for T7K; it allows T7 to infect strains that carry diverse systems but share no common one, through a brute-force deactivation strategy

Mechanistically, T7K’s C-terminal DNA-binding domain steers this activity towards DNA/RNA-binding proteins (building blocks of most defense systems). We map phosphorylation of Retron-Eco9 by T7K at critical residues; single phosphomimetic mutations at these T7K-modified sites abolish defense.

Most known phage anti-defenses proteins block a single system. T7K is different: a hyper-promiscuous, dual-specificity kinase that phosphorylates nearly all host proteins in a short, early burst in infection, with no clear sequence motif.

We found that E. coli can be a key driver of selection for a high-fitness antibiotic resistant K. pneumoniae strain in complex communities. Higher-order interactions with other community members could enhance or decrease the selective advantage of the resistant mutant.

We introduce a high-throughput method to quantify resistance-associated fitness costs inside complex gut microbiomes. This lets us identify which microbiota are permissive or restrictive for antibiotic-resistant pathogens.

Excited to share our preprint led by Carlos Voogdt et al We developed new genetic tools & genome-wide libraries for species of the Bacteroidales order; constructed saturated barcoded transposon libraries in key representatives of three genera. www.biorxiv.org/content/10.1...
