How do hundreds of proteins grip the cell’s actin skeleton? After 4 years of work, we discovered a tiny, 9-amino-acid conserved motif that acts as a universal molecular anchor—defining a whole new class of actin-binding proteins! Read it here: www.nature.com/articles/s41...

Evolutionarily conserved short linear motifs drive actin filament binding - Nature Cell Biology
Paraschiakos, Yuan et al., using a computational pipeline, identify the short linear actin filament-binding motif (SFM) as an evolutionarily conserved actin filament-binding motif present in 103 human...

All sparked by one pattern in a sequence alignment. Huge thanks to everyone involved, especially Biao, Michael, Kostja, and the Windhorst, @linderlab.bsky.social & Marlovits. Very happy to finally share the work!

Using our computational pipeline (SLiMFold), we found this motif in 103 diverse human proteins. We resolved how it engages actin using biochemistry, cell imaging & cryo-EM. Fun fact: it’s the exact same motif found in LifeAct, the widely used imaging probe!

We aligned Lifeact and F-tractin bound to F-actin and discovered a structurally conserved motif. Using this as a seed, we built a pipeline combining PSSM scoring, disorder prediction, and secondary structure filters to scan the human proteome. #Lifeact #Ftractin
