7/9 Lipoproteins had opposing effects depending on biological context: +CPPs used them to “hitchhike” into lipoprotein-dependent cells, whereas in lipoprotein-independent systems, lipoproteins acted as sponges that limited peptide toxicity.

6/9 The LDLR result led us outside the cell. Biophysical and imaging experiments showed that +CPPs bind extracellular lipoproteins, including LDL, HDL, and VLDL, and can “hitchhike” into cells through these lipoprotein-uptake pathways.

5/9 What cellular machinery controls this process? A genome-wide CRISPRi screen pinpointed the low density lipoprotein receptor !! as a major regulator of +CPP uptake.

4/9 Where do +CPPs go after entry? Using multiplexed imaging (4i), we mapped thousands of peptide-containing vesicles against the entire endolysosomal pathway with 4i and show that they escape from the lysosomes.🔬

3/9 By testing +CPPs in the natural contexts in which they evolved, from predator–prey interactions to host defense and disease, we observed endocytic uptake across human cells, tissue-derived organoids, insect cells🪰, and unicellular eukaryotes.

2/9 Whether +CPPs enter cells through active endocytosis or passive membrane “tunneling” has long been debated. Testing natural peptides from 17 species spanning ~600 million years of animal evolution, alongside disease-linked arginine-rich peptides, revealed endocytosis as the unifying mechanism!
