I had the same thought when I walked through the @ucdavis.bsky.social arboretum in 2008 and noticed displays on everything EXCEPT chemistry, so I made it part of my first NSF and now we have 5 permanent exhibits on the medicinal chemistry of natural products from plants that grow there!

Tried a few oxone conditions...ultimately after trying tons of experiments, including collaboraring with folks from here & TPFKAT (the platform formerly known as twitter), we've concluded it's just a recalcitrant substrate!!

in many cases we (and others to whom we have sent substrate) see very low mass recovery, so lots of bad things can happen!

several things...the isolable products include 3° carbon hydroxylation and quinone methide products, i.e. where the site we want to oxidize is becomes the methide carbon and the para OCH3 becomes a carbonyl.

for the top substrate, this was our runner up, i.e. we get cs. 10% product. For the bottom one, we used Ce(OTf)4 (relatively obscure paper) and we see benzylic dimerization, i.e. we isolate the product were there is a C=C bond between the benzylic sites of two substrate molecules

yeah, the e-richness causes problems, which is why we moved from the top substrate to the bottom one...more steps for the same product, but incrementally more successful chemistry...initial hope was the the double directing effect of the two OCH3 would give us regioselectivity in the top substrate

Hey #chemsky ...anyone got a super-cool benzylic oxidation that might work for this substrate? We've tried just about everything under the sun...in the top case we either see NR or consumption of SM with no product formation, and in the bottom case we top out at 25%
