USP7 sustains PAX3::FOXO1 enhancer reprogramming and represents a therapeutic vulnerability in Rhabdomyosarcoma
Rhabdomyosarcoma (RMS) is the most common soft tissue sarcoma in children and is often associated with dismal outcomes, underscoring the urgent need for new therapeutic strategies. RMS arises from emb...

PRKG1 hinders myogenic differentiation and predicts response to AKT inhibitor ipatasertib in Rhabdomyosarcoma - Nature Communications
Despite frequent AKT/mTOR pathway activation in patient’s rhabdomyosarcoma (RMS), success of AKT inhibitors in the clinical has been limited. Here, using RMS patient-derived models, the authors demons...

The new findings of Alina Ritter showcase that this approach may be especially feasible for metastatic disease since an impirtant viral docking site is highly expressed in metastatic lesions and even contributes to metastasis in #Ewing #sarcoma. We will not stop until there is a cure!

Based on the neomorphic DNA-binding capabilities of the Ewing sarcoma-specific EWSR1::ETS fusion oncoproteins, our expression constructs can deliver specifically produg convertases, (neo)-antigens, and cytokines to induce cell death only in Ewing cells, i.e. turn a 'cold' tumor 'hot'.

Since CXADR is a viral receptor, this study further paves the way for our novel virotherapy of #Ewing #sarcoma based on our 2022 study by @hoelting.bsky.social pioneering a suicde gene therapy induced by a Ewing sarcoma-specifc synthetic GGAA-enhancer tinyurl.com/2sckk89m

Neomorphic DNA-binding enables tumor-specific therapeutic gene expression in fusion-addicted childhood sarcoma - Molecular Cancer
Chimeric fusion transcription factors are oncogenic hallmarks of several devastating cancer entities including pediatric sarcomas, such as Ewing sarcoma (EwS) and alveolar rhabdomyosarcoma (ARMS). Des...
