6/ We also see close ER-mito contacts overlapping with tethers at replicating mtDNA. We propose a model, where a gradient of ATAD3A determines positioning of the fission machinery. This gradient in turn is defined by the replication state of mtDNA, reminiscent of the MinCDE gradient in bacteria.

5/ Using GEM 💎 Cryo-ET tag, we confirm that these tethers are ATAD3A. ATAD3A over-expression makes membranes more zippered, while, knockdown makes membranes floppy.

4/ In these regions, we found tethering densities that zipper 🤐 across the two membranes. These tethers cluster around replicating mtDNA.

3/ Using a CLEM-CryoET pipeline, we find that membranes around replicating mtDNA are different. There are patches of membrane which have strikingly uniform OMM-IMM distance.

2/ Osman lab described a “sphere of influence” 🔮 around each mtDNA nucleoid. We asked whether replicating mtDNA influences its local microenvironment differently than non-replicating mtDNA.

Extremely grateful to have had the opportunity to present my CryoET adventures in the grotjahn lab at #cellbio2025. We can all agree that the CryoEM and CryoET session was the best. It was wonderful to meet old mito besties, NCBS besties and make new besties. 😌
