We've updated our cleared-tissue direct-view oblique plane microscopy preprint to include our latest resolution-enhancing results. Now the Nyquist sampling of our camera is no longer a limitation and so we obtain a diffraction-limited 2 μm lateral resolution! www.biorxiv.org/content/10.6...

For an internal talk, I wanted an easy way to disprove the prevailing myth that expansion microscopy gels expand due to repulsion between charged polymer strands. I did a quick calculation of the supposed energy released during a standard experiment and it's equivalent to a small nuclear bomb...
A calculation of the energy released by four-fold swelling of a 5 mm expansion microscopy hydrogel, containing 1 mmol of sodium acrylate. The calculation assumes a sphere of uniform charge density, in water, and gives an energy of 9.4E13 Joules, or approximately 22.5 kilotons of TNT.

The final version of our multispectral microscopy manuscript is now online at Nature Photonics: www.nature.com/articles/s41... The main additions are some further comparisons to yet more unmixing algorithms, in which we find that our algorithm is still the most accurate.

We present a simple method to easily increase the imageable depth of an expansion microscopy gel on a typical inverted microscope ten-fold, using some carefully placed FEP film and a water dipping objective lens:
A comparison of imaging an expansion microscopy hydrogel with a water dipping objective lens, using either eye gel as an immersion medium or a combination of Immersol W, water and a thin FEP film. In both cases, the sample is supported by a thin FEP film rather than a coverglass.
