When KAR function is disrupted, these components become uncoupled, leading to impaired endothelial-to-hematopoietic transition and reduced blood stem cell emergence. I’m very grateful to all the collaborators and co-authors who made this work possible, and I hope you enjoy the story!

Here are a few more details and the model we think best explains the phenotype. In a functional hematopoietic niche, KAR signaling helps coordinate myeloid support, vascular organization, and hemogenic Notch signaling to promote hematopoietic stem cell (HSPC) emergence.

More broadly, this work highlights how glutamatergic signaling can regulate developmental processes far beyond the nervous system, and opens new avenues for studying how glutamate signaling regulates the function of multiple blood cell types in health and disease.

We show that KARs are required for primitive hematopoiesis and hemogenic niche organization to support hematopoietic stem and progenitor cell (HSPC) emergence. Disrupting KAR function impairs blood stem cell development, with lasting effects on adult hematopoietic clonality.

Fun fact: the original Science launched on July 3, 1880, founded by John Michels with financial backing from Thomas Alva Edison. It struggled to attract enough subscribers and stopped publishing in March 1882.

Exactly 10 years ago, I took these photos of the first volume of Nature (1869) and the “first” volume of Science (1883) at Countway Library, Harvard Medical School. It was quite a ride finding them among all the volumes in the library, but I’m so glad I did.
