We then used a completely independent strategy: genetic suppression of RAB27A, a key regulator of EV secretion. Again, EV release dropped substantially, while the senescence phenotype and soluble SASP remained intact

The result? Conditioned media depleted of EVs largely lost their ability to induce senescence in recipient cells. Fewer EVs meant less growth arrest, less SA-β-Gal activity, and lower p21 induction

Of course, antibodies should always be used with controls, but it is often difficult to find the right ones. And that's the case with those that produce bands. Some don't work at all!

What this confirms is that we researchers accept this ongoing fraud perpetrated by the companies. We all assume you will have to do a lot of research beforehand, maybe even buy several antibodies (if you can afford it), and still end up paying hundreds of euros for water.

What this reveals is a larger problem. The manipulated images in the catalogs are not the problem per se, but they expose the brazen and unscrupulous attitude of the companies selling antibodies.

You already assume those images are worthless. Actually, a long time ago I realized that many of them were duplicated and were being used to illustrate different antibodies, and I even (naively) contacted some of these companies to inform them of this "error"

Reese Richardson and Sholto David (@sholtodavid.bsky.social) have reported over 18,000 manipulated images in the antibody catalogs of 16 companies. Personally, I would never base my decision to buy an antibody on the image the company displays 🧵

Aún en shock tras leer “Medea me cantó un corrido” de la escritora mexicana Dahlia de la Cerda. Su anterior libro “Perras de reserva” me voló la cabeza y postergué leer este segundo con cierta reserva por miedo a la decepción. Nada más lejos de la realidad. ¡Qué barbaridad de relatos!
