With the Wisconsin Nathan Shock Center, we analyzed multi-tissue transcriptomic profiling data, which hinted at an essential role for liver mitochondria in the molecular response to Val-R across tissues. We validated these results, identifying Val-R induced changes in liver mitochondrial function.

We find that Val-R improves metabolic health and reduces frailty in both sexes, while specifically increasing male lifespan by 23% – suggesting that decreasing dietary valine may be a robust way to promote healthy aging and extend longevity.

We recently identified a critical role for the branched-chain amino acids – leucine, isoleucine, and valine – in regulating metabolic health. We previously showed that restriction of isoleucine by itself is sufficient to promote metabolic health and extend lifespan, but what about the other BCAAs?

Over the last decade the research of many, including our lab, as well as Drs. Luigi Fontana, Steve Simpsons and Samantha Solon-Biet, has demonstrated that a calorie is not just a calorie – and that eating less dietary protein promotes metabolic health in both mice and humans.

Surprisingly, this is not the case - ketogenesis is dispensable for the metabolic effects of CR. Indeed, Dr. Yeh discovered that CR mice adapt to the repeated fasting by CR, and over time stop inducing ketogenesis during their inter-meal fast.

CR as typically delivered leads to long periods of fasting between meals - and fasting normally induces ketogenesis. So here, Dr. Yeh tested the hypothesis that ketogenesis contributes to the metabolic benefits of CR.
