All the tested fatty acyl-CoAs fully rescued the ATP levels and partially also the synaptic function. Full rescue of the synaptic function required all three fatty acids in combination. We concluded that myristic and palmitic acid therefore likely also have non-energetic roles such as lipidation.

Why it matters? Understanding the pathway opens the possibility of biomarker-based diagnosis and metabolic therapies aimed at restoring neuronal energy supply, something that previously did not exist for this condition.

In our lab we study Hereditary Spastic Paraplegia 54 (HSP54), caused by DDHD2 mutations. We discovered that DDHD2 generates specific saturated fatty acids that fuel neuronal energy. When DDHD2 is missing, neurons are not simply structurally abnormal, they are metabolically starved. rdcu.be/eIQA0

Good question. If you read how that view came to be, it’s from studies e.g. using isolated mitochondria (excluding neuronal and Ddhd2 activity), supplementing short-chain fatty acids, or by comparing beta-ox capacity to cells with robust beta-ox such as astocytes or cardiomyocytes.
