The genetic driver of Acute Necrotizing Encephalopathy, RANBP2, regulates the inflammatory response to Influenza A virus infection
Influenza virus infections can cause severe complications such as Acute Necrotizing Encephalopathy (ANE), which is characterised by rapid onset pathological inflammation following febrile infection. H...

𝗪𝗵𝗲𝗿𝗲 𝗶𝘀 𝗠𝗜𝗧𝗗𝟭 𝗶𝗻𝗱𝘂𝗰𝗲𝗱 𝗯𝘆 𝗶𝗻𝘁𝗲𝗿𝗳𝗲𝗿𝗼𝗻? 🧠 Mouse data pointed to the 𝗯𝗿𝗮𝗶𝗻, so we tested human brain cells. Only microglia, the immune sentinels of the brain, showed interferon-driven MITD1 expression. 🚨 This makes 𝗠𝗜𝗧𝗗𝟭 𝗮 𝗺𝗶𝗰𝗿𝗼𝗴𝗹𝗶𝗮-𝘀𝗽𝗲𝗰𝗶𝗳𝗶𝗰 𝗜𝗦𝗚, protecting the brain from flaviviruses

𝗛𝗼𝘄 𝗱𝗼𝗲𝘀 𝗠𝗜𝗧𝗗𝟭 𝗯𝗹𝗼𝗰𝗸 𝘃𝗶𝗿𝗮𝗹 𝗿𝗲𝗽𝗹𝗶𝗰𝗮𝘁𝗶𝗼𝗻? 🧬 MITD1 overexpression forms cytoplasmic aggregates that deplete CHMP1B+4B, key ESCRT-III proteins. 🔬 PLA confirmed their recruitment to flavivirus replication factories. Knockdown of CHMP1B+4B hinders MITD1’s antiviral effect—proving a link

To uncover new host factors restricting flavivirus replication, we screened a library of >1,000 interferon-stimulated genes (ISGs) across multiple species. Among known antiviral ISGs, we found 𝗠𝗜𝗧𝗗𝟭, a never-described-as-antiviral-before factor, as a potent inhibitor of WNV, USUV, ZIKV & DENV
