That’s the cleaned story, but the reality was more complicated. We had to regenerate Hif1a mouse lines couple of times, the core of the story made more sense near the end of my PhD (after some “failed” projects). If you’re navigating a turbulent project and self-doubt: you’re not alone out there.

Finally, two days after viral #challenge, Hif1aiKO mice showed higher infectious titers, which were rescued by exogenous IL-21 treatment. A similar effect was observed in the survival of Hif1a iKO mice in a lung #adenocarcinoma model (10/10).

Timing mattered! Deletion at day 15 (when HIF-1α is core-enriched) decreased lung #GC -like B cells and #IgA titers, whereas deletion at day 25 (when HIF-1α shifts to the TLS periphery) mainly affected IgA titers (9/10).

IL-21 is not exclusively being sense by T or B cells! We could observe that lung #Macrophages, resident #NK cells and lung residents B cells were impaired in Hif1aiKO mice, and rescued by IL-21 (8/10).

We next asked which HIF-1α–dependent signals sustain CXCR6+ TRH cells. Intratracheal #IL-21 administration restored CXCR6+ TRH cell numbers, consistent with reduced #Il21 transcripts in sorted TRH cells and lower IL-21 levels in #BALF from Hif1aiKO mice (7/10).

Next, we generated the Hif1aiKO mouse line, which allowed us to preserve HIF-1α activity during initial priming of CD4+ T cells. Notably, lung resident CD4+ T cells were selectively decreased in Hif1aiKO mice (5/10).

Using the Egln3-YFP reporter mouse stain (generated by the Goldrath lab 🇺🇸), we tracked HIF-1α-active TRH cells. They were restricted to the lung extravascular compartment, transient during infection and localize to the borders of TLS (4/10).

This echoed earlier #scRNAseq data from the lab: lung Tfh-like resident helpers (TRH) split into two subsets highly expressing Bcl6 or HIF-1α (3/10).

Two weeks after infection, lung #TLS could be divided into inner and outer zones based on B cell proportional density. The outer zone was enriched for non-GC Tfh signatures and HIF-1α–associated pathways, such as #Hypoxia, also observed by Hypoxyprobe staining (2/10).
