A mundane question from a Friday evening brain.. If I have a protein overexpressing 20% more in a sample and it is the most abundant protein in that sample, how should I normalize the data for fold analysis for the rest of the proteins if an equal representative sample was analyzed.. #TeamMassSpec

Hello #Astral users, Have you ever come across the ion foil adjustment failure error?? We have started seeing it since the tune was upgraded to 2.2 recently during the PM. The error interferes with the system getting ready to run a sample in the step where astral mass accuracy is checked with ICS.

A question to mass spectrometrists in the core facilities around the UK and Europe.. How are you finding the cost of S-traps for proteomics applications?? It looks like incorporation of S-traps in the workflow adds an additional £8 per sample.. It feels extraordinarily expensive..

What is the magic soup in the PreOmics iST kit, I wonder.. It is getting me 6000 proteins from tiny FFPE samples!! Whereas the conventional workflow is hardly touching 1000 proteins!!

Setting up a decent DDA method on Astral is so mentally taxing.. It isn't meant to follow any of the data dependency criteria.. Enforcing it to follow the criteria is where it gets taxing.. Wonder how it is working for those MS2-TMT workflows.. 🤔🤔

Today at 13:50, on the dot, the sequences running on both Ascend and Eclipse came to an abrupt stop.. Just at random.. In the middle of a run.. Neo kept running but no MS acquisition.. Had to restart Xcalibur and reconnect LC to resume the sequence.. Very spooky!!! #MassSpecMusings

Anybody doing proteomics on FACS sorted cells? People are using BSA coated tubes to avoid cell adhesion onto the walls. BSA is all over the protein extract. How to avoid this BSA? Saw Polyvinyl alcohol (PVA) can be used in place of BSA. Any experience with its compatibility for downstream MS? Thanks

Even when the 96% of study sample contributors were females, the 4% males are the ones who get the mention!!! Wonder why can't the demography of the sample population be mentioned as 96% female or for better clarity as 96:4 female:male in the abstract.. Another instance of scientific gender bias??

Would you do tissue proteomics with an old sample fixed in formalin, not embedded in paraffin, and stored for quite a while immersed in formalin?? Can the proteins be extracted after subjecting to decrosslinking?? Are there any insights to share from the #paleoproteomics practitioners?? Thanks..
