🚨 Can DNA that does nothing become a cell’s Achilles’ heel? 🧬 Cas9 selectively killed human cells carrying a non-functional target sequence, sparing cells without it. www.sciencedirect.com/science/arti... #CRISPR #DNARepair

Weaponizing CRISPR/Cas9 for selective elimination of cells with an aberrant genome
The CRISPR/Cas9 technology is a powerful and versatile tool to disrupt genes’ functions by introducing sequence-specific DNA double-strand breaks (DSB…

🚨 Can broken DNA generate a new protein? Our study independently confirms that DSBs trigger transcription from the break—and shows, for the first time, that the resulting RNA can be translated. Could this generate neoantigens after radio/chemotherapy? www.nature.com/articles/s42...

Site-specific DNA double-strand break induces local transcription in cis and protein expression - Communications Biology
DNA double-strand breaks function as promoter-like elements that drive transcription and protein synthesis of polyadenylated RNAs from otherwise silent DNA regions.

#IPF has no cure. Median survival: 4 years. Lung transplant is the only real option. Our new paper shows this might be about to change. 🧵

www.nature.com/articles/s43... Using telomeric #antisense #oligonucleotides (tASOs)vwe restored hematopoietic #stem cell fitness, improved #vaccination responses, reduced #senescence and #inflammation, and improved the function of aged human hematopoietic stem cells. Without elongating #telomeres.

What drives the pathological consequences of #telomere dysfunction? So far elomere shortening has been considered the central issue. In our new nataging.nature.com study, we show that telomeric DNA damage signalling itself is a causal driver of hematopoietic decline. www.nature.com/articles/s43...

Aging increases both #telomeric DNA damage and #Alzheimer’s disease risk. Fabrizio d'Adda di Fagagna and coworkers now find that these processes are linked, with Aβ42 oligomers triggering ROS-mediated telomeric damage that in turn increases neurotoxicity www.embopress.org/doi/full/10....

embopress.org/doi/full/10.10… Today on World Alzheimer's Day we published in @EMBO Journal that #Alzheimer's diseases (#AD) is associated with DNA damage at #telomeres and that inhibition of telomeric DNA damage signaling with telomeric #ASO improves neurons cell viability
