New paper! A team led by @dbuendia.bsky.social and group leader Arturo Marí-Ordóñez at the GMI of the @oeaw.bsky.social investigated how plants silence potentially parasitic transposons when these “jumping genes” take over most of the genome.

Multiplexed perturbation enables scalable pooled screens - Nature Methods
This paper shows that high MOI sgRNA multiplexing maintains CRISPRi screen performance while enabling reduced cell numbers, challenging conventional reliance on low-MOI infections.

Now researchers can do more screens at a higher scale with a higher quality output in less time and less money.

We think this new approach will reshape how pooled screens are performed in settings where cells are precious, expensive, or experimentally constrained - including organoids, primary cells, and in vivo settings.

In one genome-wide cell-sorting screen, we identified regulators of ICAM-1 expression using only ~500,000 cells - a scale that would traditionally require far larger experiments.

By systematically testing multiplexed CRISPRi perturbations, we found that higher numbers of perturbations per cell can actually improve screen performance while reducing the number of cells required for genome-scale discovery.

A project from my postdoc in the McManus lab @mcmanuslab.bsky.social at UCSF @ucsfdc.bsky.social is out today 🎉 For years, pooled CRISPR screens have operated under a central assumption: one perturbation per cell. We challenged that assumption. 🤯 www.nature.com/articles/s41592-026-03095-w
