He also showcased the next-generation lung organoids containing epithelial, immune, and stromal compartments that can be maintained for up to 3 months. These models enable studies of SARS-CoV-2 infection and tissue-resident memory T cells, showcasing adaptive immune responses in organoids.

Furthermore, intestinal organoids were used to study celiac disease, revealing gliadin-induced immune activation in the stroma and providing new insights into disease mechanisms.

Particularly impressive were long-lived tumor microenvironment organoids retaining immune components, including TILs and TAMs. Fascinating data showing CD47 checkpoint inhibition increases SPP1 expression in both organoids and patients.

Daphne Bouwens from @rwth.bsky.social presented how kidney organoids can be used to study kidney fibrosis. When combined with single-cell RNA-seq atlases across disease-mimicking conditions, organoids enable identification of molecular drivers and pathways underlying fibrotic disease.

Talk by Liliana Santos-Ferreira (@i3suporto.bsky.social) on the role of glycosylation in gastric cancer. Patient-derived organoids faithfully recapitulated therapy resistance seen in patients, with fucosylation-related pathways among the most differentially expressed.

@juliabatki.bsky.social showed that extraembryonic gut cells are eliminated during mouse development through a p53-dependent cell death and are subsequently phagocytosed by embryonic gut cells. A beautiful example of how precise cellular clearance shapes embryonic development!

Ellen Fritsche from SCAHT discussed guidelines for developing New Approach Methodologies (NAMs) for toxicology testing. She highlighted neurospheres as valuable in vitro models for studying developmental neurotoxicity as they can provide mechanistic insights without relying on animal testing.

2D cultures favor pit-like cells, while 3D organoids develop glandular features with high Wnt activity. Particularly exciting were TissUse chips combining patient-derived stomach organoids with liver tissue to reveal patient-specific responses to prodrugs—a promising step toward precision medicine.

Interesting talk from @oieretxezarreta.bsky.social on the role of T cells in the gut and their contribution to colonocyte inflammation. Validation studies in organoid models showed that treatment with T-cell-derived factors disrupted epithelial integrity.
