Overview of complement effects. Complement activation generates multiple effector molecules. C3 fragments (C3b, iC3b, C3d) are covalently bound to target cell membranes. C3b on the surface of target cells can form C3 convertase, leading to further complement activation at that site. Both iC3b and C3d are ligands for 4 complement receptors (CR1-4) expressed by leukocytes and dendritic cells. C3a and C5a are soluble peptides that are released during the cleavage of C3 and C5, respectively. They are ligands for receptors expressed by leukocytes and parenchymal cells. They induce multiple downstream functions, including the chemotaxis and activation of myeloid cells to sites of inflammation. Activation can also generate C5b-9 multimers on cell membranes. C5b-9 forms a pore through the outer membrane. In sublytic quantities this can cause cell activation. If sufficient C5b-9 is formed it can cause osmotic lysis of the cell. Created with BioRender.
