↗️Trending in Chemical Science↗️ "Photoredox-catalyzed direct formylation of alkenes to access α,β-unsaturated aldehydes" by Le Liu and colleagues. 💡Broad alkene scope with late-stage aldehyde installation under visible light.💡 Read for free: doi.org/10.1039/d6sc...
Graphical abstract showing a visible-light photoredox strategy for the direct β-formylation of alkenes. A general alkene reacts with an α-chloro-N-methoxyphthalimide reagent under blue-light irradiation and a photocatalyst to generate a formyl radical, which is incorporated into the alkene to produce an α,β-unsaturated aldehyde. The method is demonstrated across more than 41 examples, delivering yields of up to 74% and E/Z selectivities of up to 95:5. Representative products include aromatic, fluorinated, highly substituted, and cyclic aldehydes. The figure highlights key challenges such as competing hydrogen atom transfer (HAT) processes and chemo- and regioselectivity, while emphasizing the advantages of easily available substrates, mild reaction conditions, broad substrate scope, and a photoredox-driven process.

Infographic showing the N-Ary Mapping Interface (NAMI) workflow for visualizing chemical space. On the left, a binary fingerprint matrix of 0s and 1s represents molecular data. An arrow points to a central panel containing coloured clusters of circles of different sizes, labelled “Global cluster centroid projections,” illustrating grouped regions of chemical space. A magnifying glass highlights one cluster, and a second arrow points to the right-hand panel, where individual molecular structures are displayed within a circle labelled “Local chemical space visualization.” The figure illustrates how NAMI identifies clusters from molecular fingerprints and enables detailed exploration of specific regions of large chemical datasets.

Have you seen this new 🔥HOT🔥 article in Chemical Science? "Universal charge-tag labelling of undetectable polymers for mass spectrometric characterization" by Jeremy E. Wulff, J. Scott McIndoe and colleagues. Read for free: 🔗 doi.org/10.1039/D6SC...
Schematic illustration of a polymer functionalized with a mass spectrometry (MS) charge tag. A blue wavy line at the top represents a polymer chain attached to a molecular structure. The molecule contains a trifluoromethyl-substituted aromatic group labeled in red as “charge-tagged hydrocarbon for MS detection,” connected through ether linkages to a triazole ring highlighted in green and labeled “robust linkage.” The triazole is linked to a phosphonium charge tag, highlighted in blue, with a PF₆⁻ counterion. Coloured boxes emphasize the triazole linkage and the phosphonium charge tag, illustrating the design of a permanently charged polymer end group for enhanced MS detection.

Schematic illustration of a molecular glue strategy targeting the Trypanosoma brucei redox protein Tpx. At the top, a trypanosome parasite is linked to a zoomed-in view of the Tpx redox cascade and a small-molecule glue structure. Three panels compare ligand-binding poses within a Tpx pocket containing a tryptophan (Trp) residue. Left: a detached ligand pose (green) fails to engage Trp, producing a weak molecular glue effect and low Tpx dimer affinity. Centre: a dynamic ligand pose (gold) shows transient interactions and intermediate glue strength. Right: an attached ligand pose (purple) forms stable interactions with Trp, generating a strong molecular glue effect and high Tpx dimer affinity. An arrow along the bottom indicates increasing chemically induced Tpx dimer affinity from left to right.

Schematic overview of a light-driven enantioselective transformation enabled by hydrogen-bond recognition. A chiral photocatalyst (catalyst 1), containing a rigid bicyclic framework and amide groups highlighted by grey boxes, interacts with a racemic aryl-substituted substrate through intramolecular hydrogen bonds (blue and magenta arrows). Under UV light (hν), catalyst 1 promotes divergent reactions depending on the substituent X. For X = OR, the reaction affords an oxindole product with an R-configured stereocentre (blue-highlighted) and up to 99.5:0.5 enantiomeric ratio (e.r.). For X = NHR, the reaction gives the corresponding S-configured product (magenta-highlighted) with up to 96:4 e.r. A note indicates that intermolecular hydrogen bonding is used for molecular recognition. A “Pick of the Week” badge appears in the lower left corner.

Figure comparing two vanadium-containing metallacyclic complexes, labelled 1-H and 1-Me. Ball-and-stick crystal structures are shown on the left, with vanadium highlighted in purple, silicon in yellow, nitrogen in blue, carbon in grey, and hydrogen in white. Simplified ring diagrams on the right identify atoms Si₁, Si₂, Cα, Cβ, and Cα′ and list selected bond lengths. For 1-H, V–Cα = 1.886(2) Å, V–Cβ = 2.069(2) Å, and V–Cα′ = 1.888(3) Å. For 1-Me, V–Cα = 1.865(3) Å, V–Cβ = 2.106(3) Å, and V–Cα′ = 1.877(3) Å. The comparison highlights differences in metal-carbon bonding within the five-membered vanadacycle rings.
