J Med Chem is on 🔥 with the editorials lately, and I highly recommend giving this one a read. Understanding free drug principles is the gateway to all of pharmacology, and is all too often mangled and misapplied by drug discovery practitioners.

Synthetic Approaches to the New Drugs Approved during 2023
This review is the next installment of an annual series that discusses the synthetic routes to access 28 small molecule drugs that were approved worldwide in 2023. A brief description of each drug’s mechanism of action, the history of its discovery and development, and the synthetic approaches published in primary or patent literature that were most likely used for clinical studies or development are included. Synthetic chemistry, used to convert complex intermediates to active compounds or build a new drug from basic building block chemicals, is critical to delivery of new drugs and treatments for disease to patients.

Many people talk about the "Golden Age of Antibiotics", but I hadn't seen it visualized properly. Just how many types of antibiotics were discovered during that time? So, I visualized it myself!
A timeline titled "The Golden Age of Antibiotics" shows when each antibiotic drug class was first available for medical use, with example antibiotics labeled. Classes are color-coded by their source: actinomycetes, other bacteria, fungi, or synthetic. Milestones include the first antibiotics (arsphenamines in 1910), as well as the discovery of many actinomycetes-derived antibiotics, such as streptomycin, and sulfonamides, penicillins, and tetracyclines.
