SUMO ligases conjugate the SUMO modifier to target proteins, thus regulating their functions. Here, the authors reveal how DNA and ATP binding by the multiple subunits of the SUMO ligase complex Smc5/6 enable its efficient SUMOylation of genome maintenance proteins.
