Now, there are over 100 types of sarcoma (!), it is an incredibly diverse family of tumors. They are all rare and ultra-rare cancers, many without any type of model that allows us to study the biology and develop effective therapies.

Over the past few years, we have developed an approach to rapidly establish and screen 3D tumor organoids. We have deployed this approach to study fast growing or indolent malignant tumors. Now we translate it to cutaneous neurofibromas.

There is no systemic therapy, and if the tumors cause any issue they need to be removed surgically for the most part. It is also a very challenging tumor type to model in the lab, as it is formed by many different cell types and abundant extracellular proteins.

Neurofibromatosis type 1 is a common inherited disorder caused by alterations in the NF1 gene. One of the most penetrant features of the disease is the formation of these benign tumors, cutaneous neurofibromas, in the dermis. They can be anywhere in the body, and numerous.

Here’s the thing about benign tumors. While they may not metastasize, they can still cause substantial health issues, particularly when they number in the hundreds, or thousands such as the case of cNFs in Neurofibromatosis type 1 patients.

People have their own motives so no judgment but personally I enjoy presenting unpublished data for early feedback

Twitter history of @soragnilab @twitter.soragnilab.com · 27/06/2023
In my experience there are two types of scientists. Those that freely present unpublished data with little to no reservation and those who don’t. Mandated policies rarely make a difference as the divide is large.

One of my favorite things about @UCLAJCCC is synergizing with other teams to make the best out of our individual expertise and develop bigger and better ways to solve big challenges in cancer - it's a great place to be! So on we go, to develop a platform to measure resistance

Now just our luck, one of the foremost experts on this method, @teitell is a close collaborator and just a couple buildings down the road! The platform they build allows us to estimate the mass of measured objects, at single organoid resolution, with time resolved measurements

The first thing we wanted to do, was find a method to monitor organoid response to therapy that was non invasive (label free preferred!), real-time and allowed us to monitor thousands of organoids at a time. The answer? High speed live cell interferometry

Have talked about all the open data and code, now time for the science! Why did we decide to develop a new, 3D organoid drug screening platform? To chase one of the biggest issues behind treatment failure and tumor recurrence: those few drug resistant cells surviving treatment 🧵

I am not discounting the effort it took to package everything and also find ways to share (for free) some of our less conventional data types - but it was the right thing to do for us

It's fantastic not to have to worry about having to keep data safe somewhere, having to find it and come up with a way to share it if someone asks, etc etc "Data available on reasonable request" is no more :)
