We then made a collection of ZF probes to label the centromeres of almost all human chromosomes. We validated them too 😎 We expect to have them ll in the coming months (5/n)

What about fluorescent dCas9? Well, upon tagging centromeres, it causes DNA damage in *up to 40% of tagged alleles*, and triggers detectable aneuploidy after 10d 💔 This doesn’t happen with ZFs, which label just as good! (4/n)
gamma-H2AX Immunofluorescence image + quantification showing that fluorescent dCas9 probes trigger DNA dammage when targeting centromeres, while Zinc Finger probes do not

ZATELLITE uses fluorescently-labeled ZFs designed with an AI model to target and label any DNA sequence. One plasmid, one transfection, and repetitive loci are readily visible as fluorescent foci ✨ (3/n)
cartoon showing the principle of ZATELLITE, and representative fluorescence image of cell nuclei with different copy numbers for chromosome 7 showing the matching number of foci for the ZATELLITE probes.

Sharing ZATELLITE 🛰️ - a tool to visualize and manipulate repetitive loci in living cells using designer zinc fingers tinyurl.com/ZATELLITE led by Nestor Saiz, collab with Marcus Noyes, @teresadavoli.bsky.social, @glennislogsdon.bsky.social, @liamholt.bsky.social & @jefboeke.bsky.social (1/n)
principle of ZATELLITE: synthetic Zinc Fingers are fused to a fluorescent protein and targeted to repetitive endogenous sequences - this ensures damage-free tagging, which is not the case with dCas9

I looked up the name of my upcoming study section in the register, and only saw the entries for the previous cycles. Which leads me to infer that it'll likely not happen next week www.federalregister.gov/documents/se...

after *cough cough* one or two failed attempts at cacio e pepe, I was just thinking to myself I needed to systematically explore the phase diagram to find the sauce sweet spot... scooped!

great read- struck by the similarities with this piece on spotify. "a song stays on repeat not because it’s our new favorite but because it’s just pleasant enough to ignore." 🙃 www.newyorker.com/magazine/202...

Instead, we found that the endonuclease of the Integrator complex, which terminates transcription on numerous genes, is required for the formation of Pol II clusters 4/6

📣 New preprint! Pol II forms clusters, some of which are giant: Pol II clusters at fly histone genes could fill a yeast nucleus!👇How do these structures regulate transcription? This is what Feiyue Lu, a joint postdoc w Ruth Lehmann, looked into 1/6 biorxiv.org/content/10.1...
